About the dart thing, they have to be within ten meters of the mosasaur in order for the dart to have enough force behind it to penetrate it's four inch thick skin. They mention this around the 20/21 minute mark. If they are too far away the dart would still hit it, but it wouldn't penetrate deep enough to get the sample. Also shooting it through the water would decrease the force propelling the dart which would again mean it wouldn't go deep enough be able to collect the blood needed. Shooting it while it's jumping out of the water might work to hit it and have the needle penetrate deep enough, but when the mosasaur crashed back into the water it would likely dislodge the dart and they'd be left without a sample because the dart wouldn't be full so the rocket wouldn't activate. Which is why it took her so long to take the shot, also they probably have a limited supply of darts so they wouldn't want to use more than absolutely necessary.
