doosik wasn't supposed to shoot her he was supposed to shoot the informants that's why they guy was yelling shoot the informants are getting away. he was like their backup in case anything went wrong but he also failed.

And, the gun lady which was the lady that owned the hair saloon, she has the ability to see through an object, that's why she's able to shoot her target even if something is blocking. She shot Amazon seeing through the wall, she tried to shoot doosik and can see through him by seeing through the trees that were blocking him. The agent that captured doosik at the end was the one in charge of the mission of killing the north Korea informant and he was asking the "eagle" (doosik) to shoot the people running away but doosik couldn't shoot, that's why here he told him "u screwed me again".
