PLUS it could be an unintentional show of dominance by Seina as it's her alpha genes that make her bite back in the first. It also shows that Isao recognizes that Seina's pheromones heightening is making her begin to spiral from both the head injury and pheromones, and her stubbornness is making it worse so grounding her in this way is his way of caring for her as he's always done!
