The reason L is looking to see if Light can determine the Second Kira theory is that if Light *was* Kira, Light might not want L to think there are two Kiras. That way Light could direct L and the investigation team into investigating the second Kira, and throw them off the fact that they were kind of onto him. It would also show L that Light might not be being truthful in general, since L was extremely confident that Light would realize that there was a second Kira from the tape. If Light didn't say anything, L would guess that Light did figure it out and wasn't saying it for some reason. However, just because Light says that there's a second Kira doesn't mean that any of the work that L and the team did before is wrong, just that this particular point doesn't implicate Light. So if Light says there's a second Kira, he remains as suspicious as before, but if he doesn't, it means he's probably hiding something and he gets more suspicious.
