So much stuff was cut from the manga that lead Near to the conclusion Mikami could be X-Kira. When Demegawa was first killed, Mikami was unsure whether he had Kira's blessing to choose the next spokesperson so he went on Sakura TV again and gave the speech where he says "I'd like to hear Kira's voice and thoughts once more. I think that following Kira's teachings exactly as ordered is the quickest path to world peace." At 10:24 in your reaction happened in the period of time after Demegawa was killed and before Takeda was chosen as the next spokesperson. Basically it was Mikami's coded message to Kira saying that he would pick the next spokesperson if he didn't hear anything from Kira soon. Takeda was a relatively inexperienced anchor woman compared to a lot of choices for the role, so Near figured she was chosen because X-Kira must have known she personally sympathized with Kira's cause. He then proceeded to go through ALL of Takeda's media appearances prior to being chosen as spokesperson. It was then that he stumbled across her two debates with Mikami that she moderated. Near noticed Mikami from the recent Kira's Kingdom broadcast, and with the context of that episode airing right before she was chosen, Near recognized his words for what they were, a secret message for Kira.

Yes, there is actually a replica of the Eiffel Tower in Tokyo, called "Tokyo Tower" and ironically it is even taller than the original Eiffel Tower! :D

They cut out a ton of stuff. Mogi is a much more important character as well in the manga

I dont like the misa takada scene either. Also its a better scene in the manga. Also youre right a lot was cut from the manga to the anime for this last arc. Its very unfortunate

I believe they did cut out some moments with Near that would make his assumptions more valid
