Just follow her instructions exactly in the earlier quests (don't kill if she don't ask you and so on). Make sure you have at least one xxx scene with her before you get into the Asylum. Also, after the Arkham assault event make sure Harley always stays in your hideout to get bonuse scene in the end of the chapter, which will improve your relationship as well.
