The core of the Homunculi is a phiosoper stone. In order to kill them you have to destroy the stone by completely running it out of energy. Causing any kind of damage to them will force the stone to activate and heal them so while it doesn't have to be fatal wounds as "life for a life", it's impossible to know exactly how much energy a life is worth or how many were used to create it in the first place. Though it's not required the reason fire has proven so extra effective is simply a matter of constant damage over time. While the body is burning the stone is also expending energy to maintain/restore it's condition, but once that enegy runs out good night. It may not seem like it but EVERY time we've seen a homunculus take major damage it was absolutely gettng them closer to death we just don't know how close they actually were. As for Sloth him being the fastest makes a kind of contrarian sense. While yes he's purely lazy he wants to get everythng he has to do done as fast as possible so he can just stop.

The reason Sloth is fast despite being Sloth is because Sloth (The Sin) isn't "slowness" (unlike Sloths the animal, which are named that because they're legit slow, which is a bit confusing lol). Sloth is laziness (basically, it's a bit deeper than just that, going into things such as apathy and self neglect etc, but that's the gist). So Sloth (The Homunculus) can actually move that fast usually, he isn't slow - he's just really lazy and doesn't care to move that fast most of the time. He's not bothered to put effort into anything, let alone moving fast lol.

They use the souls inside of them, when they use their alchemy. Those souls are limited in number. Regeneration takes energy. So it's limited.

#Sunday Rundown So the reason Sloth died was because he used up all of the energy from his Philosopher Stone. While the stone seem like they have limitless energy, they do indeed have a limit.

I think you guys missed a bit of foreshadowing around the time Yoki Al and the rest were arguing about the car being stuck...
