There is no real logic behind why Cersei immediatly suspected him other than her irrational hatred of him . I don't disagree that suspecting him is just absurd since the circumstances of him being the cup bearer was not something HE intended to happen and as even Curtis(other reactors have called this out too) pointed out, everything he did was in plain view of everyone else and he inspected the goblet as if he were confused on how poison got into it. He was even concerned when Joffrey was coughing as if to suggest "You good, dood?"

Everyone has talked in-depth about how Rob and Ned made mistakes that cost them their lives, but now we have Joffrey. He's a young megalomaniac who only wants to play with his subjects, doesn't listen to anyone, and has no interest in helping the very people he keeps in power. He was going to die. People like Cersei, Tywin, and Ramsay have gotten this far because they aren't idiots. There are exceptions to this, some fail upwards, some learn quickly, some do everything right and still end up dying. By the way, Rob's wedding was called the Red Wedding and Joffrey's wedding was called the Blue Wedding.
