I think Cap knew that Tony's parents were killed by Hydra from the headlines video the A.I. version of Armin Zola showed him and Natasha in the bunker in Captain America: Winter Solider.

Original movie with Christopher Lambert, Sean Connery, and a very young Virginia Madison. Fun film, great soundtrack by Queen, and lots of emotion! Hope you check it out.
