As far as the "proof of divinity" thing - Chloe tells Lucifer in season 1 that she doesn't believe in the Bible stuff, hence her telling Lucifer in this episode that she doesn't accept that he's the Devil. The conversation Chloe has with Ella about "if you could prove it was all real - would you" is one of the reasons she decides not to test the blood. What Amenadiel is talking about is physical proof of divinity - in season 1 when Lucifer's wings are stolen we get to see how they affected the people who came in contact with them. The blood would cause a similar effect (I believe) :)
