No that's not her sister. The ghost was Makomo and Shinbou's sister is Kanae. Makomo was just one of Urokudaki's other students. Kanae was a Hashira. And we've seen her twice. First time was when Shinobu and Tanjiro had their first talk on the roof. We saw a flashback of Kanae dying in Shinobu's arms. Then in the episode with Kanao's backstory we saw her again.

Some people here are speculating that Shinobu is talking about Tamayo being the demon that killed her sister but I don't think so. Tamayo has been free from Muzan's control and not harming humans for a long ass time. Shinobu enlisting Kanao's help implies that the demon is someone she can't beat on her own and the Hashira have been shown to be able to easily kill the lower moons. Also her sister was a Hashira as well. It makes the most sense that she's talking about an upper moon especially as we are apparently headed into the last arc and still have 3 of them left to deal with. So my guess is one of the final battles will be Shinobu and Kanao vs either Upper 1, Douma, or Akaza. More than likely Upper 1 or Douma because I'm sure they're gonna have Tanjiro vs Akaza.

It's crazy how Giyu thinks he's unworthy of being a Hashira when he became a Hashira at 15 years old. Giyu is so good it only took him 2 years to kill an upper rank demon. Not to mention he created his own form of breathing "Dead Calm." He's one of the strongest out of all of the Hashiras yet he still feels he's not enough.
